DNA Purification and Sorting
ZHIXUAN DNA Clean Beads are based on the SPRI principle(Solid Phase Reverse Immobilization), and are suitable for DNA purification and fragment size sorting in high-throughput sequencing library construction. ZHIXUAN DNA Clean Beads are compatible with DNA and RNA library construction kits of various commonly-seen brands and with the library construction processes mentioned in research literature. They are used in exactly the same way as the widely used AMPure XP Beads. The output and size distribution of the library are highly consistent with AMPure XP Beads. Therefore, they can seamlessly replace AMPure XP Beads and effectively reduce the library construction costs.

1. Accurate Sorting through two-step magnetic bead purification
The DNA library is constructed using TruePrep DNA Library Prep Kit V2 for Illumina (#TD501). The initial library size is 200-1500 bp. The PCR products are sorted using ZHIXUAN DNA Clean Beads according to the conditions in below table, to obtain libraries of different sizes, which were then analyzed by Agilent 2100 Bioanalyzer.
2. DNA Library Construction, Compared with AMPure XP Beads
Taking 50 ng human DNA as the initial template, construct libraries using TruePrep DNA Library Prep Kit V2 for Illumina (#TD501). Use ZHIXUAN DNA Clean Beads and AMPure XP Beads respectively to sort the libraries of an average length of approximately 470,570,670bp (corresponding insert size 350,450,550bp) under the same conditions, which are then analyzed using an Agilent 2100 Bioanalyzer.
3. RNA Library Construction, Compared with AMPure XP Beads
Taking 1 μg or 100 ng of Universal Human Reference RNA (UHRR) as the initial template, use TruSeq RNA Sample Prep Kit V2 (Illumina #RS-122-2001) to construct RNA libraries. Use ZHIXUAN DNA Clean Beads and AMPure XP Beads under the same conditions to obtain libraries of an average length of approximately 280bp (corresponding insert size of approximately 160bp), which are then analyzed using an Agilent Bioanalyzer.

FAQ
Q1: If the magnetic beads are accidentally placed at -20°C environment but not frozen, can they still be used?
A1: Not recommended for use. Because low temperature will destroy the structure of magnetic beads, reducing their capturing efficiency. It's necessary to purchase new magnetic beads for use.
Q2: What is the principle of magnetic bead sorting?
A2: In the first round, the magnetic beads bind to DNA with a larger molecular weight, and this part of the product is removed by discarding the magnetic beads. In the second round, the magnetic beads bind to DNA with a larger molecular weight in the remaining product, and DNA with a smaller molecular weight is removed by discarding the supernatant.
Many components in the initial sample will interfere with the effect of two-round magnetic bead sorting. Therefore, when the sorting scheme is executed at different positions, the amount of two-round magnetic beads used also varies.
Q3: Can single-stranded DNA be purified using magnetic beads? If so, how many magnetic beads should be used?
A3: Magnetic beads can purify double-stranded DNA and single-stranded DNA. Please refer to the instruction for double-stranded DNA for the multiples of magnetic bead purification.
Q4: Can N411 be used to extract DNA?
A4: Using N411, you can directly lyse the sample and extract gDNA by using magnetic beads. However, the efficiency may not be as high as that of specialized kits, because our magnetic beads are specially dedicated for purification and sorting.
