ZHIXUAN Universal Circularization Kit for MGI is a modular kit specially optimized for MGI high-throughput sequencing platform. The universal MGI platform circularization module provided in this kit is compatible with both single-end (Single Barcode) and paired-end (Dual Barcode) adapter type libraries, and can be used to prepare a single-stranded circular DNA library specially for MGI high-throughput sequencer. All reagents provided in the kit have undergone strict quality control and functional validation to ensure the stability and repeatability of library construction.

The circularization experiments of Zhixuan #NM202 is compared with Zhixuan #NM201 and similar products from other suppliers, Supplier A ~ D. The results show that Zhixuan #NM202 had certain advantages in the effective circularization efficiency of single-end and paired-end libraries, and that Zhixuan #NM202 is compatible with different library input amounts (0.1 pmol - 2 pmol) and different template types.

FAQ
Q: Abnormal yield of circularization product.
A:
1) The quantification of Input DNA (MGI platform linear dsDNA library) is inaccurate, resulting in low or high input amount. It is recommended that the initial input amount be controlled within 0.5 - 2 pmol, and the recommended input amount is 1 pmol.
2) Inaccurate size of DNA main fragment (presence of adapter dimer, over-amplification of large fragments, broad peaks, etc.) can lead to low or high DNA input (MGI platform linear dsDNA library). It is recommended that the library peak shape be controlled within an appropriate range.
3) Enzyme activity and function have been affected due to improper storage of reagents. It is recommended to perform the circularization again with problem-free reagents.
4) The quantification of the circularization product is not accurate. It is recommended to recalibrate the instrument before each quantification, and then take 2 μl of the circularization product and 198 μl of Qubit (Invitrogen #Q10212) reagent, vortex and mix them well before conduct quantification.
5) Operation problems, such as imcomplete mixing of reagents and reaction system. It is recommended to vortex the buffer and Oligo to mix until clear and uniform, and the enzyme can be mixed by lightly shaking or vortexing at a low speed. After each step, pipette or vortex at medium or low speeds before setting the sample onto the equipment. The reaction system in the PCR tube shall be clear and uniform, and bubbles shall be avoided.
